Generation of Gene-Engineered Human Hepatoma Cells with Heat-Inducible Liver Functions

Hepatoma cells derived from liver carcinoma are a candidate cell source for bioartificial (BAL) systems due to their high proliferative capacity, although function of hepatoma is considerably low compared with primary hepatocytes. In our previous study, genetically engineered mouse inducible were established by transducing liver-enriched transcription factor (LETF) genes. this we aimed develop new gene-engineered human cells, in which functions heat treatment. For purpose, constructed gene expression system eight LETF genes under control tetracycline-dependent transactivator (tTA), and the was introduced into genome HepG2-HSP tTA induced heat-shock protein promoter transcriptional amplification HepG2 cells. Thus, heat-inducible promotes induce function. Upon treatment (HepG2-HSP/8F) at 43°C 30 min, such as albumin secretion cytochrome P450 significantly enhanced. The can be used various hepatic studies including construction BAL systems.
